Can I let out one room to a new tenant on a joint tenancy

I will try and keep this as simple as possible. I have a HMO with 6 tenants, 2 of the tenants are on a joint tenancy agreement letting out two rooms. One of them has given notice, the other hasn't. I am issuing s21 to the remaining tenant. My question is can I legally relet the vacant room while the other tenant is living in the house (until I get possession) even though it is on the joint tenancy agreement?
